MK-677 Research Overview
MK-677 is an orally active growth hormone secretagogue that has been found to significantly increase growth hormone (GH) and insulin-like growth factor 1 (IGF-1) levels in healthy older adults without serious adverse effects. It has been associated with increased fat-free mass (FFM), body cell mass, and body weight, while not significantly affecting abdominal visceral fat or total fat mass. MK-677 also showed potential in increasing markers of bone formation and resorption, suggesting it could have implications for bone health.
MK-677 (Ibutamoren) and Growth Hormone
MK-677, also known as Ibutamoren, is an orally active growth hormone secretagogue that stimulates the release of growth hormone (GH) in the body. It has been shown to increase GH and insulin-like growth factor 1 (IGF-1) levels in both young and older adults [R, R].
In a study with healthy elderly subjects, daily oral administration of MK-677 for two weeks increased GH concentrations in a dose-dependent manner, with a 25 mg/day dose increasing the mean 24-hour GH concentration by 97%. However, the long-term effects of MK-677 on somatic growth in rats were not observed, despite the GH stimulatory effect of MK-677 [R].
MK-677 and Bone Health
MK-677 has been shown to have a positive impact on bone health. In a study with healthy postmenopausal women, daily oral administration of MK-677 for six weeks increased bone mineral content and bone mineral density at the lumbar spine and femoral neck. Another study found that MK-677 administration in rats increased the width of the tibia growth plate, although it did not increase body growth or serum levels of IGF-I [R, R].
MK-677 and Sleep Quality
MK-677 has been shown to improve sleep quality in both young and older adults. In a study with healthy young subjects, high-dose MK-677 treatment resulted in an approximately 50% increase in the duration of stage IV sleep and a more than 20% increase in REM sleep compared to placebo. n older adults, treatment with MK-677 was associated with a nearly 50% increase in REM sleep and a decrease in REM latency [R].
These findings suggest that MK-677 may improve sleep quality and potentially correct the relative hyposomatotropism of senescence.
MK-677 and Skin Health
There is limited research on the direct effects of MK-677 on skin health. However, since MK-677 increases GH and IGF-1 levels, it may indirectly contribute to skin health. GH and IGF-1 play essential roles in skin homeostasis, wound healing, and collagen synthesis.
